Two-Photon Holographic Mesoscope Probes Activity Across Large Brain Regions

BERKELEY, Calif., Dec.18, 2023 — Recent advancements in two-photon (2P) mesoscopes have extended the field of view (FOV) for deep brain imaging, making it possible to image brain regions as large as 25 mm2. Until now, though, mesoscopes could only measure neural signals; they could not stimulate separated regions of the brain. To enable investigators to probe the inter-areal communication signals central to brain function, a team at the University of California, Berkeley (UC Berkeley) combined its aeroPULSE FS50 ultrafast fiber laser for photostimulation with a 2P mesoscope from Thorlabs Inc. that has a 5- × 5-mm...Read full article

    optogenetics
A discipline that combines optics and genetics to enable the use of light to stimulate and control cells in living tissue, typically neurons, which have been genetically modified to respond to light. Only the cells that have been modified to include light-sensitive proteins will be under control of the light. The ability to selectively target cells gives researchers precise control. Using light to control the excitation, inhibition and signaling pathways of specific cells or groups of...
    holography
Holography is a technique used to capture and reconstruct three-dimensional images using the principles of interference and diffraction of light. Unlike conventional photography, which records only the intensity of light, holography records both the intensity and phase information of light waves scattered from an object. This allows the faithful reproduction of the object's three-dimensional structure, including its depth, shape, and texture. The process of holography typically involves the...
