The light source for widefield fluorescence microscopy might seem like an unlikely enabler for high-speed live cell imaging, but the level of control that comes with solid-state LED Illumination Systems has changed the game.
Digital control via TTL capitalises on the maximum speed of LED switching, but often requires expensive specialist components. The CoolLED pE-300
ultra Illumination System (or the and pE-340
fura for calcium imaging) overcome this with two innovate features, enabling the capture of dynamic cellular processes with speeds of up to 10 µs. The ability to fit inline single-band excitation filters allows for a Pinkel configuration, removing the cost and latency of a filter wheel. The CoolLED Sequence Runner simplifies TTL control by utilising the TTL output signal available on most scientific cameras and cycles though LEDs in a user-selected sequence for each TTL signal.
This simple approach provides affordable high-speed microsecond switching precisely synchronised to the camera exposure, which also enhances data accuracy through minimised phototoxicity and photobleaching.
