GaAsP Nondescanned Photodetector

Olympus Europa Holding GmbH has released the FV10MP-BXD-GAP photodetector for enhanced sensitivity when performing multiphoton fluorescence imaging.

The nondescanned detector captures very faint fluorescence signals and minimizes the laser power required to generate a detectable signal, making it suitable for use in advanced life sciences research. High sensitivity is achieved by coating the photoelectric surface of the detector with GaAsP, which offers better quantum efficiency than conventional photomultipler tubes (PMTs), the company says. Peltier cooling keeps noise to a minimum, boosting the signal-to-noise ratio.

The detector includes standard PMTs for identifying regions of interest (ROIs), which can then be scanned using the GaAsP PMTs, maximizing their lifetime. When researchers use the photodetector in combination with an Olympus FluoView FV1000MPE multiphoton microscope system, they can produce insightful images, even when using advanced irradiation techniques, low laser power or very faint fluorophores.

The photodetector is designed to make use of every photon emitted by a sample. Efficient photon detection means that a wide range of biological processes can be investigated using fluorescently tagged molecules expressed at physiologically relevant concentrations, even at levels previously too low to be detected using traditional multialkali PMTs. Because a lower intensity of laser light is required to accurately detect each fluorophore, the power of the laser can be reduced, minimizing the effects of photobleaching and phototoxicity, and increasing the accuracy and biological relevance of results, especially for experiments performed over long periods or requiring frequent laser light exposure.

The device is equipped with two conventional multialkali PMTs for routine browsing of samples, and two GaAsP PMTs for highly sensitive analysis of a specific ROI. Switching between the two types is fast and easy. To lengthen the lifetime of the GaAsP PMT, the system prevents unnecessary exposure to ambient light such as room lighting or excessive fluorescence.
